To unlock most gladiators in arena mode, simply defeat them. Exceptions to this are listed below.
Narcissus/Slave
Beat him in the Atilius Arena ( Rank 2 or higher.)
Narcissus/Gladiator
Beat him in the Atilius Arena ( Rank 3 or higher.)
Commodus/Gladiator
Lose to him in the Colosseum. ( Rank 2 or higher.)
Commodus/Emperor
Defeat him during the event on day 48. ( Rank 3 or higher.)
Laetus
Defeat him during the event on day 50. ( Rank 3 or higher.)
